I believe this is what you should do.

This means that the bindings are expecting to find the entire boost tree
somewhere rather than just a set of installed libraries and headers.

I don't think you have to.

OK that narrows it down somewhat.
I don't know about boost.build on Windows but are you sure it is finding
your user-config.jam? The error message suggests it does not know the
rule python.get-python-interpreter. On Linux I have user-config.jam in
my home directory. You seem to have put it at $(BOOST_ROOT)\build\v2\
